If I setup a new account on Amazon using a new computer tethered to an android phone which, in turn, will be linked to a router being used by another account, will Amazon know?
Basically, I want to run two accounts from one router connection so, if the phone uses wifi from the router then connects to the computer using it's hotspot facility, how will this appear to Amazon ?

Your question does not make any sense. Your phone will use it's 3G/4G data as a hotspot, not the wifi from your router, which means it will have a different IP address compared to the router.
Anyways, be more clear next time. Maybe you will get a better answer.
