can we use highlight box in main picture when we listing the item?

Do you mean colours around the edge etc? - no, ebay policy stipulates your pictures must be absolutely clean of graphics. I have had listings taken down for thumbnails not conforming to this request.

A little bit, such as a UK flag in the corner, I have seen people get away with.
