The Dymo 450 is for little address labels, not 4x6 shipping labels.
Invest in a Dymo 4XL Labelwriter, buy your 4x6 labels from Amazon from a listing with a lot of good reviews (there are some crappy labels for the 4xl on there.)
Having a Stamps.com account (USPS) is free if you go through Shipstation. You can link it to your preferred credit card (rewards card is good if you're doing a lot of $$ in shipping.) If you're UK, idk how it works.
Make sure your packaging will work for 4x6 labels. You can wrap the tracking code over the side of a box if the box is too short to fit the label on one side, but don't wrap any of the return/to address portion unless you want them coming back to you.
My big problem is getting the labels that shoot out of the printer onto the packages in a time-effective manner. Shipstation gives you the option to print several things as part of the label (i.e. order number) that may help do them in assembly line fashion. I'm still working on optimizing the labeling process.
