I think you should be more worried about being limited... But you should wait at least 12-24 hours, yes.

What if I get more money in while I'm waiting the 12 hours? Should I wait an additional 12 hours, or just do a partial withdrawl?

The big question is why are you depleting your paypal funds as quickly as you get them. Sounds like some kind of scam. I am surprised paypal hasn't limited you already.

Double posting is annoying and unnecessary

I would think pp would expect people to withdraw most of their balance to bank acct after they'd sold their items, especially if they sell a batch of items once every two weeks for example instead of continuous selling. Assuming all payments from a batch of items have come into pp, how long would you recommend waiting before transferring all/part of the balance?

I keep a $1,000 USD running balance to cover charge-backs, fees, etc. Then again that is probably why my PayPal accounts have been in good standing for the past 4 years it isn't like I try to take the money and run. 4 accounts all in good standing. Patience is a virtue that is what you have to have with the eBay and PayPal rules.
